Abstract:
The first section of this thesis briefly describes the chemistry of tetracycline (A) and related compounds. A short account of the published syntheses of tetracyclines is included. This is followed by a summary of the progress made in these laboratories towards the synthesis or tetracycline. Attention is Focused on the work relevant to the author's contribution. The second section contains a discussion of the author's work directed towards the synthesis of tetracycline. It is- divided into three parts. Part I describes the synthesis of the tetracyclic intermediate (B). Part II relates to the synthesis of a different tetracyclic intermediate (C) and contains a subsection dealing with experiments conducted on the photochemical step of this synthesis. The last part is concerned with experiments directed towards the oxidation of phenols to o-hydroxycyclohexadienones, e.g. (D) to (E). The emphasis is on reactions which could be directly applied to the 12a-hydroxylation of (C) and thus be useful in the synthesis of tetracycline.
