Abstract:
The techniques of steady state and flash-induced oxygen evolution, flash-induced absorption spectroscopy and chlorophyll fluorescence spectroscopy have been used to study two aspects of the light reactions of photosynthesis in the unicellular green alga Ghlorella emersonli. (1) The effects of various compounds have been investigated in order to deidve more information in elucidating the electrogenic mechanism responsible for the slow phase of the flash-induced absorption change at Sub-uncoupling concentrations (circa 0,8muM) of carbonyl cyanide-m-chlorophenyl hydrazone (CGGP) have been found to stimulate the apparent extent of the slow phase of 515 and markedly decrease the rate of decay in Chlorella but not in chromatophores of Rhodopseudomonas capsulata or in intact or osmotically shocked chloroplasts isolated from Pea and Spinach. Similar results were observed using the other uncouplers carbonyl cyanide-p-trifluoromethoxyphenyl hydrazone (FGGP) and Z,k dinitrophenol (DNP), and the plastocyanin inhibitors, KGN and HgGl2. (2) The effects of GuSO4, HgCl2, ZnSO4 and Pb(NO3)2 have been investigated in order further to chaxacterise the inodes of toxicity of these heavy metal ions on photosynthesis. Cu2+ has been found to inhibit steady state oxygen evolution at approximately 10.
