Abstract:
Numerous studies have shown that transplantation of mesenchymal stromal cells (MS Cs) for the treatment of ischemic cardiovascular disorders improves tissue function through paracrine mechanisms. However. the injected cells are gradually lost at the site of injury, thus liming the window of therapeutic efficacy. In this project, after a detailed comparative study of MSC isolation procedures and therapeutic potential in vivo in the mouse, we screened for novel factors improving bone marrow-derived MSC engraftment and long-term survival. Exploiting an innovative Functional Selection (FunSel) procedure developed in our laboratory, we selected protective factors mediating cell retention in the ischemic myocardium directly in vivo. The most effective molecule was cardiotrophin 1 (Ctfl), a pro-hypertrophic cytokine never associated with this property before. We deeply investigated the effect of Ctfl on MSCs and demonstrated that this factor is a powerful cytokine promoting cell adhesion in the ischemic myocardium thus prolonging the beneficial effect of cell transplantation after cardiac damage.
