Abstract:
Poly(ADP-ribose) Polymerase-1 (PARP-1) is an abundant nuclear enzyme involved in the repair of DNA single-strand breaks via the Base Excision Repair (BER) pathway. Cemozolomide, an alkylating chemotherapy agent with activity in melanoma and gliomas, causes DNA methylation and single-strand breaks that can be repaired by BER. Pre-clinical studies have shown that small-molecule PARP inhibitors can potentiate temozolomide-induced cytotoxicity and anti-tumour activity in vitro and in vivo.
