Abstract:
This thesis presents the results of investigations into the synthesis, characterisation and biological behaviour of some radiophannaceutically-relevant copper, zinc and rhenium complexes. Chapter One gives a brief introduction into the uses of radiation in medicine. Emphasis is placed on complexes of rhenium, technetium and copper. Specific attention is paid to the importance of (thiosemicarbazonato)copper(II) complexes as hypoxia-selective PETimaging agents. Chapter Two presents the synthesis of a range of simple zinc thiosemicarbazone complexes, which are structurally analogous to some medically relevant copper thiosemicarbazones. The photophysics and cytotoxicity of all the complexes are reported. In addition, the first fluorescence measurements of the uptake and distribution of thiosemicarbazone complexes in living cells are presented, for [Zn(ATS!vI)] and [ZnCI(ATSPyrrH)]. Chapter Three describes the synthesis and characterisation of a number of fluorescent copper thiosemicarbazone complexes. The cytotoxicity of the complexes is also given. Chapter Four introduces a number of criteria upon which copper(II) complexes can be evaluated for hypoxia-selectivity. The synthesis of a number of copper complexes is described and their suitability for hypoxia-selectivity based on these criteria is given. Results of further studies on the most promising candidates are also detailed. Chapter Five contains a brief synopsis of the synthesis and photophysical characteristics of a range of tricarbonyl rhenium(I) complexes, which have potential for. use as radiotherapeutic agents. Results from investigations into the uptake and distribution of these complexes via fluorescence microscopy are given. Chapter Six gives the experimental details and characterising data for the preceding Chapters Two - Five. In addition, the methods used in the biological studies are also described. Appendices A - D contain details of the single crystal X-ray structure determinations.
