Abstract:
Psoriasis is a common chronic inflammatory skin disease affecting 2-3% of the Caucasian population. Although the aetiology of the disease remains poorly understood, there is strong evidence that psoriasis has a significant genetic predisposition. This study investigated the genetics of Type 1 psoriasis using a number of approaches to prioritise candidates for investigation including: a gene-focussed fine mapping study of the linkage peak at the susceptibility locus PS0RS9 (PDGFC, GLRB, GRIA2, IL15 and TIR2) extended to include two closely related genes of potential biological significance (TLR4 and TLR6 with TLRl); an investigation of a gene already associated with a number of related diseases (FTPN22); and, a validation study of novel association reported in a GWAS of psoriasis patients (IL12B and IL23R).
